Abstract

The kinetics of enzymatic hydroxylation of cortisol at C-2α and C-6β by liver homogenate preparations has been studied in 2 genetic guinea pig types (strain 13 and specially bred Hartley) distinguished by high and low production of cortisol hydroxylated derivatives (high and low producers). Conditions were established under which linear progress curves obtained, allowing the determination of initial rates. The enzymatic hydoxylation resided primarily in the microsomes and no significant activity was found in the soluble (100,000 ×g supernatant) fraction. Optimal pH and TPNH concentrations and the relationship between enzymatic rates and tissue concentration were determined with microsomal preparations consisting of the 5000 ×g supernatant. A fair linear relationship between enzymatic rates of cortisol 2α- and 6β-hydroxylation and tissue concentration was observed up to 20–30 mg/ml with both strains.
