Abstract

A sensitive high performance liquid chromatographic method with fluorometric postcolumn labeling using 2-cyanoacetamide was developed for determination of unsaturated oligosaccharides produced from hyaluronic acid (HA) by digestion with Streptomyces Hyaluronidase. The unsaturated tetra-and hexasaccharides produced enzymatically from HA were separated completely within 30 min by HPLC. The detection limit was 10 ng of HA. This method was applied to the determination of HA in normal rabbit plasma to verify the sensitivity and practicability.
