Abstract
Objective To develop a method of quantitative analysis of multi-components by single marker (QAMS) to measure five effective components in Sangju-Ganmao grannules. Methods Withthe HPLC method, the chromatogram column was Agilent Zorbax SB-C18 (250 mm × 4.6 mm, 5 μm), the mobile phase was acetonitrile -0.2% physcion acid in gradient elution program, and the flow rate was 1.0 ml/min. The detection wavelength was set at 340 nm for chlorogenic acid and rutin, 205 nm for phillyrin, 254 nm for quercetin and glycyrrhizic acid, the column temperature was 30 ℃ and the injection volume was 10 μl. Using chlorogenic acid as the internal standard to measure the relative correction factor of four components like rutin, phillyrin, quercetin and glycyrrhizic acid by QAMS and calculate their content, and compare the cnents with the external standard method. Results The calculated values of chlorogenic acid, rutin, phillyrin, quercetin and glycyrrhizic acid were not statistical significant from those measured by external standard method (P>0.05). Conclusions The QAMS method can be used for the measurement of the content of five effective components in Sangju-Ganmao granules and the method is simple, effective and accurate.
