Abstract

The study describes the development of a method for determination of ethylenediaminetetraacetic acid (EDTA) in various biological matrices after extraction as sample preparation using liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). The method is based on the complex formation of the excess of EDTA with FeIII. The excretion of EDTA in swine, treated with high doses of EDTA against dysentery, was monitored over a week by measuring its concentration in urine, plasma, and feces after stopping the administration of the medication. After slaughter, the EDTA concentration was measured also in the fatty tissue, kidney, liver, and muscle in order to determine the distribution of its residue between the tissues. The new method is fast, and the one-step sample preparation procedure may be regarded as derivatization. The method is applicable for low-concentration analysis with 3.0 ng mL−1 limit of detection and 10.0 ng mL−1 limit of quantification, while the relative standard deviation does not exceed 4.3 %.
