Abstract

A simple, sensitive, and accurate HPLC-CL method was developed for the first time to determine 2-undecanone in the dried aerial parts of Houttuynia cordata Thunb (HC), which has been widely used as one of the more common traditional Chinese medicines (TCMs). This HPLC-CL assay was performed on a reversed-phase C18 column (250 mm × 4.6 mm, 5 µm) at the column temperature of 35°C. The mobile phase was composed of acetonitrile and water (50:50, v/v). The flow rate was 1.0 mL/min. The chemiluminescence was produced by luminol oxidation during a reactor of which was based on the analytes enhancement effect on the CL reaction between luminol and K3Fe(CN)6. The calibration curves showed good linear regression (r 2 = 0.9993) within the test range of 3.0 × 10−8−1.0 × 10−5 g/mL. The established method showed good precision with average intra-day and inter-day variations of 2.08% and 1.85%, respectively, and a recovery of 99.55% for the 2-undecanone. The validated method was successfully applied for the determination of 2-undecanone in 9 samples from 9 different cultivation regions. The present HPLC–CL method was demonstrated to be very helpful in the quantification of 2-undecanone in HC.
