Abstract

Coliform detection using Lab on Chip Devices (μPADs) are effective yet economical alternatives to conventional methods. The current work involves standardisation of coliform detection using β-galactosidase (β-gal) assay on LOC devices using Chlorophenol red-β-Dgalactopyranoside (CPRG) as a substrate. The assay was standardised using positive control culture E. coli ATCC 25922. As proof of concept studies, the device was tested using spiked food samples. This was followed by testing of the devices with actual samples for presence of coliforms and the results obtained were compared with results obtained by traditional methods. The limit of detection of this device was found to be 2.0xrentity×103CFUmL-1.
