Abstract

During 2013 and 2014, nearly 10 % of violet orychophragmus plants in Nanjing, China, exhibited typical phytoplasma-infected yellow dwarf symptoms. The pathogen was identified using nested-PCR amplification through phytoplasma-specific universal primers. Phytoplasma 16S rDNA and rp gene fragments were successfully amplified and sequenced in the diseased plants. Phylogenetic analysis based on 16S rDNA and the rp gene indicated that violet orychophragmus yellow dwarf phytoplasma was related to ‘Candidatus Phytoplasma ziziphi’ (16SrV-B), which was also verified by virtual RFLP analysis results of 16S rDNA. This is the first report of ‘Ca. Phytoplasma ziziphi’ infecting violet orychophragmus with yellow dwarf symptoms in China.
