Abstract

An assay was developed for the characterization of a tobacco rattle virus (TRV) isolate from potato tubers exhibiting disease symptoms resembling corky ringspot disease. Using reverse transcription and the polymerase chain reaction. cDNA of this TRV isolate was obtained for the 16 kDa open reading frame located on RNA-1. Amplified cDNA was cloned and sequenced to confirm the identity of the virus and to determine the relationship of this isolate to other sequenced TRV isolates. Disease symptoms and virus morphology were also examined and used to characterize this Canadian TRV isolate. Deduced amino acid sequence of the recovered TRV isolate had a high level of homology (97.9%) with the PLB isolate of TRV from The Netherlands.
