Abstract

A new library of 1,2,3-triazole incorporated 1,3,4-oxadiazole-quinazoline derivatives (11a-j) were prepared and characterized by 1HNMR, 13CNMR and mass spectroscopic techniques. Further, these synthesized compounds were evaluated for their anticancer effects on four human cancer cell lines such as PC3 (prostate cancer), A549 (lung cancer), MCF-7 (breast cancer) and A2780 (ovarian cancer) by a MTT assay and etoposide used as positive control. Most the tested compounds displayed good anticancer activities as positive control. Among them, compound 11a, 11b, 11h and 11i demonstrated more potent anticancer properties on four cell lines. Particularly, one compound 11a exhibited highest anticancer activity. Further, Molecular docking studies were carried out for all 1,2,3-Triazole Incorporated 1,3,4-Oxadiazole-quinazoline Derivatives 11(a-j) against hDHFR (PDB ID:1U72). Compound 11a with 3,4,5-trimethoxy substitution on the phenyl ring has shown highest binding affinity ΔG = −11.65 kcal/mol.
