Abstract

:Objective To observecytotoxicity of in vitro expanded NK cells in a panel of malignant glioma cells andinvestigate potentials of adoptive NK cell therapy for patients with malignant gliomas.Methods NK cells and lymphokine -activated killer(LAK) cells were obtained from PBMC of 4healthy individuals and expanded with KRATM media. Cells proliferation was measured bycell counting and specific cell markers in-cluding CD3/CD16/CD56 on NK cells wereconfirmed by flow cytometry. MTT assays were employed in cy-totoxicity assays. Results Thein vitro expansion rate of NK cells was up to 7.93 × 109 in total, the cytotox-icity ismore effective with NK cells than that of LAK cells against all tested glioma cells,including LN18 LN229 、T98G and U87MG. Conclusion NK cells expanded from PBMC in vitro are effectivekilling cells a-gainst malignant glioma cells, this may imply their anti - cancerpotential for adoptive cell therapy with ma-lignant gliomas.
