Abstract
We have developed a method for distinguishing between α-N- acylaminoacyl-tRNA and unblocked α-aminoacyl-tRNA. This procedure depends on catalysis of ester hydrolysis of α-aminoacyl-tRNA by Cu 2+ at pH 5.5 or 7.5. Acylation of the α-amino nitrogen group of the aminoacyl ester results in loss of sensitivity to the action of Cu 2+. Previous treatment with Cu 2+ does not impair the capacity of the tRNA, subsequently precipitated with ethanol in the presence of EDTA, to accept amino acids.
