Abstract
A new method of estimating amylase by the cup is reported. The stainless steel cups, which we employed have the external diameter of 8mm, the internal diameter of 6mm, and the height of 10mm, and are the same as those now commonly used for penicillin cup bioassay. Enzyme solutions were filled in each 5-6 cups in a petri dish of the diameter of 90mm, containing 5cc of 0.1% soluble starch in 2% agar solution and incubated for 18 hours at 45°C. After incubation, cups were took off and iodine solution was poured into dish. Colourless clear rings were for med and their diameters were measured. In the definite concentration range of diastase, such as 0.5g_??_4.0g in 100cc distilled water, we could found the simple equation between clear ring diameter (D), LINTNER units (L) and amylase concentration (E).
