Abstract

[Co(L)(H2O)2]ClO4 (where L denotes N,N′-Bis(3-methoxysalicylidene)-1,2-diaminopropane) was prepared, and its characterization was reported by single crystal X-ray crystallography, elemental analysis, mass spectrometry and spectroscopic methods (1H NMR, IR, and UV–Vis). The crystal structure showed that the arrangement around cobalt atom is octahedral with two imine nitrogen atoms (N1, N2) and two oxygen atoms of a hydroxyl group (O2, O3) in equatorial positions and two water molecules at the axial positions. The hydrogen bonding interactions between a coordinated water molecule and phenoxo/methoxy oxygens result in supermolecular dimers. Interaction of the complex with FS-DNA and protein (BSA) was explored by absorption and fluorescence spectroscopy. Moreover, Cyclic voltammetry (CV), viscosity measurements, circular dichroism (CD) and agarose gel electrophoresis were applied to determine the binding affinity to DNA. According to Kb value (≈102), the complex is moderately bound to DNA. The complex interacts with BSA by both static and dynamic quenching. Results of molecular docking were used to obtain detailed binding information of the complex to DNA and BSA. Cytotoxic activity was performed by MTT assay. It was observed that the complex exhibited low toxicity against the MCF-7 and LNCaP cancer cells.
