Activation of the mTORC1/PGC-1 axis promotes mitochondrial biogenesis and induces cellular senescence in the lung epithelium. Am J Physiol Lung Cell Mol Physiol 316: L1049–L1060, 2019. First published March 20, 2019; doi:10.1152/ajplung.00244.2018.—In the above article, Fig. 2A (p-mTOR and mTOR), Fig. 2B (PGC-1α), Fig. 4D (NRF-1), Fig. 10B (p-p53), and the related densitometry calculations were published incorrectly. Below are the corrected figures and data. The figure legends remain the same, and the authors assert that the corrections do not affect the conclusions of the figures or the study. Figure 2.The mammalian target of rapamycin/peroxisome proliferator-activated receptor-γ complex 1 (mTOR/PGC-1) axis is upregulated in alveolar epithelial type II (AE2) cells from the lungs of old mice. A: Western blot (WB) for phosphorylated (p-)mTORC1Ser2448, mTORC1, p-p70 S6Thr389, and p70 S6 in AE2 cells from young and old mice. B and C: WB for PGC-1α, PGC-1β, nuclear respiratory factor 1 (NRF1), and transcription factor A, mitochondrial (TFAM) in AE2 cells isolated from lungs of young and old mice. D: WB for Tom20 in AE2 cells. E: representative transmission electronic microscopy of AE2 cells from lungs of young and old mice. Quantification of number of mitochondria per AE2 cell is depicted in bar graph. F: mitochondrial (mt)DNA/nuclear (g)DNA ratio in AE2 cells (n = 6, each group) isolated from young and old mice. G: quantification of reactive oxygen species levels by MitoSox staining of AE2 cells isolated from young and old mice. Western blot images are representative of 2 different blots, and results of densitometry analysis are depicted in bar graphs (n = 6, per group). Statistical significance was assessed by Student’s t-test. *P < 0.05, **P < 0.01, ***P < 0.001 vs. control group.Download figureDownload PowerPointFigure 4.The mammalian target of rapamycin/peroxisome proliferator-activated receptor-γ complex 1 (mTOR/PGC-1) axis is upregulated in senescent (Sen) L2 cells. A: Western blot (WB) for phosphorylated (p-)mTORSer2448, mTOR, p-p70 S6Thr389, p70 S6, and β-actin in control and senescent L2 cells. B: transcript levels for Pgc1α and Pgc1β in control and senescent L2 cells. C: WB for PGC-1α and PGC-1β in control and senescent L2 cells. D: WB for nuclear respiratory factor 1 (NRF1) and transcription factor A, mitochondrial (TFAM) in control and senescent L2 cells. Densitometry is shown on the right. Statistical significance was assessed by Student t-test. *P < 0.05, **P < 0.01, ***P < 0.001 vs. control group.Download figureDownload PowerPointFigure 10.MitoTempo treatment reduces cellular senescence (Sen) markers in bleomycin (Bleo)-exposed rat L2 cells. A: representative images of β-galactosidase (β-Gal) activity (blue cytoplasmic staining) in control, L2+BLM, L2+MitoTempo (Mito), and L2+BLM+Mito groups. Percentages of β-Gal-stained cells are shown on the right. B: Western blot (WB) for phosphorylated (p-)p53Ser15, p21, p53, and β-actin in control, L2+BLM, L2+Mito, and L2+BLM+Mito groups. C: transcript levels for Il6, Mcp1 (monocyte chemoattractant protein-1), Tnfα (tumor necrosis factor-α), and Il1α in control, L2+BLM, L2+Mito, and L2+BLM+Mito groups. D: WB for p-p70 S6T389, p70 S6, and β-actin in L2+BLM+pRK7, L2+BLM+pRK7+Mito, L2+BLM+S6K1, and L2+BLM+S6K1+Mito groups. E: WB for p21 and β-actin in L2+BLM+pRK7, L2+BLM+pRK7+Mito, L2+BLM+S6K1, and L2+BLM+S6K1+Mito groups. Images are representative of 2 different blots, and results of densitometry analysis are depicted in bar graphs (n = 6, per group). Statistical significance was assessed by ANOVA. **P < 0.01, ***P < 0.001 vs. senescence group.Download figureDownload PowerPointThis article has no references to display.
