Abstract

Objective To investigate the association of varying polymorphisms of zinc finger protein-36 (ZFP36) and obesity in Xinjiang Uygur population.Methods A total of 932 Uygur individuals were enrolled in the study with random clustered sampling for assessment of the genotypes.Following screening of the mutant points of ZFP36 gene by sequencing in 48 Uygur individuals with obesity,the representative points were selected for subsequent genotyping and case-control study by using Taqman-PCR on the basis of an extended population.Results The difference in distribution of individuals with obesity,overweight and normal controls,based on body mass index (BMI),did not reach statistical significance for genotypes CC,CT and TT and alleles C and T corresponding to rs3746083 of ZFP36 gene (P>0.05).No marked difference in the distribution of three genotypes (P>0.05),but not that of the alleles (x2=14.314,P<0.05),was noted in individuals with abdominal obesity and normal waist circumference on the basis of waist circumference.The distribution of frequency of genotypes CC,CT and TT and alleles C and T for rs3746083 differed considerably between males with obesity and normal waist circumference (x2=6.264,x2=8.307,P<0.05,respectively),which was absent in females (P>0.05).The genotype profiles suggested that the differences in fasting blood glucose and insulin resistance for rs3746083 between those with genotypes CT+ TT and CC were statistically significant (P<0.05).When grouped on the basis of BMI,the genotype distribution frequency of AA,AG and GG and alleles A and G for rs251864 in individuals with obesity,overweight and normal controls did not appear statistically significant (P>0.05).There was no profound distinction in the genotype distribution frequency of all genotypes and alleles for rs251864,on the basis of waist circumference,in individuals with abdominal obesity and normal waist circumference (P>0.05).The AA,AG and GG genotypes,but not the corresponding alleles,yielded a marked difference in rs251864 between males with abdominal obesity and normal waist circumference (P<0.05).Nonetheless,this distinction in females with abdominal obesity and normal waist circumference did not reach statistical significance (P>0.05).There was a marked difference in the diastolic blood pressure among patients with genotypes AA,AG and GG of rs251864 (P<0.05).Conclusion The zinc-finger protein-36 rs3746083 polymorphisms may be associated with abdominal obesity in Uygur males.
