Abstract

Objective To investigate inosine monophosphate dehydrogenase (IMPDH) gene polymorphisms involved in mycophenolic acid (MPA) response and their association with acute rejection one year after renal transplantation.Methods We genotyped three single nucleotide polymorphisms (SNPs) in 105 Chinese Han renal transplant patients.Genotyping was performed using derect sequencing SNPs Genotyping Assays.The association of SNPs with 1-year post-transplantat acute rejection was analyzed.Results Of the 105 patients,the IMPDH1 rs2278293 genotype frequencies of GG,GA and AA were 33.33%,46.67% and 20.00% respectively.The IMPDH1 rs2278294 genotype frequencies of GG,GA and AA were 20.95%,46.67%,and 32.38% respectively.The IMPDH2 rs11706052 genotype frequencies of GA and AA were 13.33% and 86.67% respectively.The IMPDH1 rs2278293 and rs2278294 were associated with acute rejection over the first year post-transplant.Combined rs2278293 and rs2278294 genotype was associated with acute rejection one year after renal transplant.Conclusion IMPDH1 rs2278293,and rs2278294 SNPs were associated with a lower risk of acute rejection and may improve MPA treatment outcome.
