Abstract
Objective To discuss the application effect of different antigen retrieval methods with autoclave and microwave oven in immunohistochemical staining. Methods 12 cases of esophageal cancer tissue specimens in our hospital from August 2016 to June 2017 were collected for the study, four different antigens (CD4, CD8, Ki67, IL17) located in the cell membrane, nucleus, cytoplasm were selected to do staining, two antigen retrieval methods with autoclave and microwave oven were performed. The immunohistochemical staining results of the two antigen retrieval methods were analyzed. Results The antibody positive rate of CD4 of the antigen retrieval method with autoclave was as high as 100%, which was higher than that of the antigen retrieval method with microwave oven (45.5%) (P 0.05). Conclusion In the case of non-shedding tissue, the antigen retrieval method with autoclave has the advantages such as simple operation, light background coloring, clear positioning, stable result; in the case of no significant difference in the positive rate of retrieval cases, the microwave antigen retrieval method is proposed.
