Mice were treated with vehicle (PBS) or simvastatin (1 mg/kg) IP, and after 24 hours the mice were anesthetized and injected with rhodamine 6G to label leukocytes. The mesentery was exteriorized, superfused with 20 μL histamine 1 mmol/L, and rolling leukocytes were imaged with a fluorescent digital camera. B, Quantification of simvastatin inhibition of leukocyte adhesion in mice (n=3 to 5±SEM. *P<0.04 vs vehicle). C, Simvastatin increased S-nitrosylation of NSF in vivo. Wild-type or eNOS knockout mice were treated with simvastatin for 24 hours, and spleen lysates were immunoprecipitated with antibody to nitrosocysteine. Precipitants were fractionated and immunoblotted with antibody to NSF (upper) to detect nitrosylated NSF (NSF-NO). To confirm that simvastatin did not change total NSF expression, total cell lysates were also immunoblotted with antibody to NSF (lower).
