Abstract

Since publication of our work [Cytoskeleton 69(5): 324–335, 2012; doi: 10.1002/cm.21029], we learned that the anti-MYH7B antibody generated against the synthetic peptide QRHLERALEERRRQEE (corresponding to residues 1457–1472 of the mouse MYH7B sequence) is not specific for this myosin isoform. While our affinity-purified antibodies do indeed detect MYH7B protein, they also detect MYH6 protein. Note that the peptide sequence used as antigen in preparation of the antibody does not appear in the MYH6 sequence. We wish to thank Drs. Leslie Leinwand and Alberto Rossi for making us aware of this cross reactivity and for (1) sharing their anti-MYH7B-specific antibodies that do not detect MYH6 (alpha-cardiac) in mouse heart muscle lysates (data not shown) and (2) for testing our antibodies on human GFP-MYH7B and rat mCherry-MYH6 (alpha-cardiac) that were transfected into HEK293 cells; both demonstrated reactivity by Western blot analysis which indicate that our anti-MYH7B antibodies are not isoform-specific and recognize MYH6 (Correction, Fig. 1). p. 327: “MYH7B protein is expressed in the mouse heart and assembles into myofilaments.” p. 322: “To demonstrate that protein was indeed synthesized from MYH7B transcripts, we raised antibodies against a unique sequence of mammalian MYH7B and used this to detect a single robust band in mouse heart and extraocular muscle tissue (Fig. 4).” We regret any confusion that our antibody studies may have caused.1 Anti-MYH7B antibody recognizes MYH6. HEK293 cells were transfected with constructions encoding an EGFPHuman MYH7B fusion protein (GFP-Hum MYH7B) or an mCherry MYH6 fusion protein (mCherry Rat MYH6). Protein blots of extracts from these cells indicate that the anti-MYH7B antibody described in our work, recognizes MYH6 protein.
