Abstract

A new spectrophotometric method for the determination of proto-, copro- and uroporphyrins is described. It is based on the formation of copper or zinc chelates from porphyrin methyl esters, using metal acetate solution in chloroform/methanol under conditions which have been defined for quantitative conversion. The calculations are made on the basis of the molar absorptivity in the Soret bands. Extinction coefficients are presented. The analysis is specific and more sensitive than the corresponding measurement of the metal-free porphyrin esters in chloroform solution. For this reason the method is especially suitable for spectrophotometric analysis of submicrogram quantities of porphyrins.
