Abstract

Continuous flow fast atom bombardment (CF-FAB) mass spectrometry provides a means for particle bombardment analysis of aqueous solutions continuously infused into a mass spectrometer. Flow-injection analysis of biological reactions or systems are readily accommodated by this technique. This chapter describes the basic concepts and performance advantages of CF-FAB as compared to normal (standard) FABMS. Many specific applications are discussed, including direct injection of peptide mixtures obtained from protease digests. The coupling of CF-FAB for applications involving on-line liquid chromatography and capillary electrophoresis is described in some detail and is illustrated by the analysis of tryptic digest of proteins. Finally, the use of CF-FAB with microdialysis is described for the in vivo analysis of drugs and endogenous metabolites. In these applications, mass specific detection is used in drug pharmokinetic analysis and in the monitoring of the time-course of drugs crossing the blood/brain barrier. In conclusion, the outlook of the technique is briefly discussed.
