Abstract
In most respects, preparation of maize genomic libraries in lambda phage is similar to preparing any other genomic library. We follow the procedures for cloning with lambda phage vectors detailed by Sambrook et al. (1989) with the variations described below. We focus here on those aspects that may require special attention when using maize DNA. Please note the “Other Considerations” before beginning to clone.
