Abstract

Subunits of glutenin were identified by their apparent molecular weight (MW) determined by 17·3% polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulphate (SDS-PAGE). The method was applied to estimate the MWs of the 24 high MW (HMW) subunits identified numerically on the basis of gene product analysis. MWs obtained with 10% polyacrylamide gels were generally about 15% lower than the values obtained with 17·3% gels. For the four subunits for which the amino acid sequence has been published; the 17·3% gel values were generally 32 to 54% higher than the actual values. For these four subunits, MW values determined by amino acid sequencing and by 17·3% gel were linearly related with an r 2 value of 0·98.
