Abstract

PIEZO proteins are homotrimeric cation-selective mechanosensitive ion channels that transduce various forms of mechanical forces into intracellular biochemical signals. Using multi-scale molecular dynamics simulations, we have generated an open state of the PIEOZ1 channel from the cryo-EM structure of a nonconducting conformation that reproduces unitary ionic conductance and selectivity in wild-type and mutants channels (Jiang, et al.
