By deconstructing the current two-cell model of embryonic lymph node development in mice, they placed lymphatic endothelial cells (LECs) as key initiators and regulators of lymph node formation. We agree that a more prominent role for LECs within this model is valid; however, we posit that the proposed latter model, in which LECs determine location and control of lymph node initiation, is not supported by the literature and needs reconsideration. The current model describing LN formation in mice is based on a two cell-type interaction scheme with lymphoid tissue inducer cells regulating the activation of mesenchymal lymphoid tissue organizer cells. We highlight here the key role of lymphatic endothelial cells during the initiation of LN formation. The involvement of lymphatic endothelial cells as an additional organizer cell type in LN organogenesis unveils multiple control levels that govern the generation of lymphoid organs.
