Abstract

The vanadium(V)-thrombin complex was prepared in solution by the interaction of H2VO4 − with human thrombin. The interaction between vanadate and the protein was characterized by UV spectroscopy and titrations. The binding constant was calculated to be 104−−9 M-2. A model complex of vanadium with trypsin was also prepared and studied by UV difference spectra to compare the binding sites of trypsin with thrombin for vanadate. It is concluded that vanadium is binding as vanadate to carboxyamide groups of the glutamine residues.
