Abstract

The efficacy of several extractants for the release of ATP from soil samples was compared. This comparison was based on (1) the quantity of ATP extracted and (2) the adenylate energy charge determined when bacterial cells were added to the soil samples. The most ATP was extracted from soil and either an equivalent or higher adenylate energy charge was obtained using an extractant composed of phosphoric acid, urea, dimethyl sulfoxide, adenosine, ethylenediaminetetraacetic acid and Lubrol. This extractant was a modification of a previously described one (Webster, J.J., Hampton, G.J. and Leach, F.R. (1984) ATP in soil: a new extractant and extraction procedure. Soil Biol. Biochem. 16, 342–355) in which Zwittergent 3,10 was replaced by the less expensive detergent Lubrol.
