Abstract

Enzyme activities involved in carrot phytoalexin 6-methoxymellein biosynthesis were determined by an improved assay method. Irrespective of the elicitor such as a metal ion, ethylene or oligogalacturonides, 6-methoxymellein production took place at almost the same rate as the O-methyl group transfer reaction to a hydroxyl group of 6-hydroxymellein, a direct precursor of 6-methoxymellein. Synthetic activity of 6-hydroxymellein was ca two-to four-fold that of the O-methyltransferase. These results suggest that O-methylation of the dihydroisocoumarin precursor is a rate limiting step of 6-methoxymellein biosynthesis.
