Abstract
The lactogenic activity of baboon, sheep, chinchilla, hamster, rat, mouse, rabbit, guinea pig, and dog placentae at various stages of pregnancy was examined by organ coculture of placental explants with prelactating mammary tissue, as well as by addition of placental extracts to cultured mammary tissue. Mammary tissue from pregnant (days 11–12) BALB/cCrgl mice was cultured in Waymouth's synthetic medium supplemented with insulin (5 μg/ml) and aldosterone (1 μg/ml). The lactogenic response to placental explants and extracts was judged histologically by comparison with the response to ovine prolactin (5 μg/ml). Placental lactogenic activity of baboon, sheep, chinchilla, hamster, rat, mouse, and guinea pig was clearly demonstrable. In the rabbit and dog, activity was not detectable. Although placental endocrine differences exist among the Mammalia, the variety of species examined to date is insufficient to allow any evolutionary generalization.
