Abstract

Aim: Lysosomal acid lipase (LAL), coded by LIPA gene, has critical role in cholesterol-ester metabolism in LDL receptor pathway. LIPA is a susceptibility gene for CAD in GWAS studies, as LIPA SNP with increased expression shows increased CAD risk. LAL activates SREBP and LXR, and LAL deficiency shows non-HDL overproduction and HDL reduction. LAL showed lipotoxicity in pancreas ß-cells. However, little is known about physiological LAL activity.
