Abstract

Petiole protoplasts and leaf mesophyll cells were enzymatically isolated from the sweet potato cultivars ‘Jewel’ and ‘Centennial’. When cells or protoplasts were plated in the appropriate media formulations, colony formation was readily observed. Protoplast or mesophyll cell derived colonies could continue proliferation to callus on a range of secondary media.
