Clive2, Zsolt Liposits1 and Imre Kalló1* 1 Hungarian Academy of Sciences, Hungary 2 King’s College London, United Kingdom 3 NRA/CNRS/University of Tours/Haras Nationaux, France Besides the classical neurotransmitters, neuropeptides also play significant roles in the regulation of gonadotropin releasing hormone (GnRH) secretion. Galanin and neurokinin B (NKB) have a modulatory function on GnRH secretion. In contrast, kisspeptin (KP) initiates a long-lasting activation of GnRH neurons. KP neurons in the rostral periventricular area of the third ventricle (RP3V) have been shown recently to convey estrogens’ positive feed-back action on GnRH secretion, whereas kisspeptin neurons located in the arcuate nucleus (Arc) have been implicated in the negative feed-back effects of estrogen. The aim of this study was to determine, whether galanin or NKB are co-expressed with kisspeptin in the mouse RP3V and/or Arc or alternatively, they are synthesized in different subsets of cells. Dual-label immunohistochemistry for KP and preproneurokinin B or galanin was carried out on sections from colchicine-treated female mice representing the full rostrocaudal extent of either the RP3V or the Arc. Maximum kisspeptin expression was detected in the RP3V of sections originating from ovariectomised (OVX) and estrogen-treated females, whereas the Arc showed the highest levels in sections derived from OVX females. Confocal microscopic analysis revealed that almost all KP neurons in the Arc, but none in the RP3V express NKB. In contrast, galanin immunoreactivity was observed in nearly every KP neurons in the RP3V, whereas in the Arc galanin immunoreactivity was detected only in 15% of KP neurons. These observations raise intriguing questions about the physiological roles of these neuropeptides potentially co-released in the vicinity of GnRH neurons.
