Abstract
Simple methods are described for the collection from sheep of polymorphs or macrophages suitable for use in in vitro studies. Very large numbers of polymorphs (150 to 500 millions), virtually free from other types of cell or contaminant material, were harvested from the involuted mammary gland 4 to 12 hr. after the infusion of 5 μg. of endotoxin in saline. The washings obtained 6 days after the infusion of endotoxin contained 110 to 190 million cells, of which about 75% were macrophages. A smaller number of macrophages were harvested from glands following the infusion of paraffin oil. The majority of the macrophages obtained after infusion of paraffin oil were vacuolated, whereas most of those obtained after infusion of endotoxin were non-vacuolated.
