Abstract
The behaviour of chicken liver xanthine dehydrogenase (xanthine: NAD +—oxidoreductase (E.C. 1.2.1.37)) with oxygen or with NAD + as electron acceptors is compared. With oxygen, the double reciprocal plot of v versus [xanthine] 0.5 is linear, whereas for NAD + the 1/ v versus 1/[xanthine] plots are linear. With NAD + as substrate (xanthine:NAD +—oxidoreductase activity or with NADH (diaphorasic activity)), the double reciprocal plots of v versus [substrate] are linear. The pattern of uric acid inhibition of the xanthine dehydrogenase against xanthine varies with the electron acceptor considered. The kinetic constants characterising the systems have been determined.
