Abstract

YiiP is a zinc/proton antiporter in the Cation Diffusion Facilitator superfamily. YiiP contributes to zinc homoeostasis by coupling zinc export to the proton motive force. Zinc binds at three sites: Site A in the transmembrane domain (TMD) constitutes the transport site. Site B is in a poorly conserved cytoplasmic intracellular loop connecting transmembrane helices M2 and M3. Site C binds two zinc ions in the cytoplasmic, C-terminal domain (CTD). Previous cryo-EM analysis revealed a novel conformational change featuring a kink between the TMD and the CTD, bending of M2 and M5 helices, and closure of a hydrophobic gate hypothesized to control the accessibility of the transport site.
