Abstract

Identification and molecular analysis of high molecular weight glutenin subunits (HMW-GS) of wild emmer wheat T. dicoccoides K5199 were carried out. Novel HMW glutenin subunits 1AxTd and 1ByTd were identified. Nucleotide sequences of 1AxTd and 1ByTd genes were detected (GenBank accession numbers MH475136 and MG897125, respectively). The coding sequence of 1AxTd was similar to 1Ax1 of T. aestivum (99.7%), while the 1ByTd gene sequence was similar to the genes of other T. dicoccoides samples (98.5 and 97.6%). Primary and secondary protein structures of the studied HMW-GS allowed for predicting a high contribution of the 1AxTd subunit to the breadmaking qualities and the average contribution of 1ByTd thereto. Evaluation of the most important grain quality traits identified increased protein and gluten content as well as reduced rheological properties of T. dicoccoides K5199 gluten.
