Abstract
Guinea pig mitogenic factors (MF's)† appear to be a heterogeneous group of molecules which vary in mol. wt and isoelectric point according to the stimulus used for their production. Antigen-induced MF's are basic proteins which have mol. wt 20,000–25,000. Lymphocytes stimulated with insoluble concanavalin A (iCon A), on the other hand, produce two major MF activities which have mol. wt 40,000–50,000 and acidic pI values. iCon A-activated lymphocyte supernatants contain proteolytic enzyme activity to 3H-acetyl casein which co-purifies with mitogenic activity. Pretreatment with the serine esterase inhibitor phenylmethylsulphonyl fluoride (PMSF) suppresses protease activity and mitogenic activity of one fraction of MF (pI 4.4–4.6) by 75%. The other MF (pI 5.4–5.6) was inhibited by 20–30%. Skin reactive factor (SRF) activity is also found in one fraction of MF activity (pI 4.4–4.6). This fraction no longer causes skin induration if it is preincubated with PMSF. Skin reactivity, therefore, appears to be related to proteolytic activity.
