Abstract
For over 35 years, high-performance anion-exchange chromatography with pulsed amperometric detection (HPAE-PAD) has been used to determine a wide variety of carbohydrates in an even wider variety of samples. This chapter provides the analyst with information pertinent to successfully choosing, installing, running, and maintaining the proper liquid chromatography system for HPAE-PAD carbohydrate analysis. This chapter also discusses how to successfully execute four common HPAE-PAD carbohydrate analysis applications and an application that should be used with greater frequency in the coming years. The four common applications reviewed are monosaccharide analysis of glycoproteins and similar samples, sialic acid analysis of glycoproteins and similar samples, carbohydrate analysis of samples used for biofuel production, and glycoprotein oligosaccharide profiling. The fifth application is the hyphenation of HPAE-PAD oligosaccharide profiling to mass spectrometry.
