Abstract
The emergence of the drug design and combinatorial chemistry has led to the emergence of drug candidates of high molecular weight, with poor water solubility and high lipophilicity. The pharmaceutical formulator faces the challenge of new drug development primarily due to poor water solubility of the drug. Poor solubility can lead to poor bioavailability manifesting in suboptimal drug delivery. The advent of various insoluble drug delivery technologies has facilitated formulation of poorly water soluble drugs into an optimal drug delivery system. Several marketed drugs have been reformulated to improve efficacy, safety and patient compliance. Revitalization of poorly soluble drugs using improved drug delivery technologies have been successfully adopted by many pharmaceutical companies to gain marketing exclusivity and patent protection for such products. This chapter covers the conventional techniques and recent advances in the field of poorly water soluble drug delivery.
