Publisher Summary This chapter describes the recent work designed to evaluate the specific neuroendocrine response capabilities of male and female hypogonadal mice with preoptic area (POA) grafts (HPG/POA), and neuroanatomical studies that explore some possible factors involved in directed gonadotropin hormone-releasing hormone (GnRH) outgrowth to the median eminence. The neuroendocrine studies include the evaluation of plasma luteinizing hormone (LH) pulsatility and of the ability to show alterations in LH release in response to acute or chronic changes in the gonadal hormone milieu. Related to this estrogen-concentrating cells present within the grafts and in the host hypogonadal brain. The chapter describes the tracing methods, neurotoxic lesions, the identification of tanycytes and glia, and co-grafts in neuroanatomical studies. Future studies will attempt to identify differences in afferents to the grafted GnRH cells (or to their axonal projections) that may underlie specific neuroendocrine capabilities.
