Abstract

Objective To study the cytokines level in the early stage of rats with septic shock treated by different types of fluid resuscitation. Methods One hundred and fourteen SD rats were randomly assigned to 6 groups: control group, septic shock group, sham operation group, physiological saline recovery group, low molecular dextran recovery group, and human albumin recovery group.Septic shock rat model was made by using cecum ligation puncture method (CLP), and the blood was collected respectively on postoperation after 6 h, 12 h, 24 h to detect the cytokines level.The concentrations of TNF-α, IL-6, IL-1β, IL-10, IL-4 in serum were detected by means of enzyme-linked immunosorbent assay (ELISA). Results Compared with control group, TNF-α, IL-6, IL-1β and IL-10 level were elevated in all time point (all P 0.05); Compared with shock group, the level of IL-4 (anti-inflammatory factor ) in all the liquid recovery group was higher than that in the shock group in 24 h time point(P 0.05). Compared with the shock group, the 3 liquid recovery groups had higher survive rate. Conclusions Fluid resuscitation may restrain the cytokines outbreak in early stage in septic shock and decrease mortality through reducing the levels of TNF-α, IL-6 and IL-1β of rats and increasing the levels of IL-4 and IL-10.
