Abstract

Swelling of Brassica napus leaf protoplasts occurred in both a Murashige and Skoog, (MS) medium and an ammonium nitrate-free MS medium, (MS (-NH4+)) medium. The swelling was also observed in both light and dark cultures. A gradual decrease in the concentration of soluble protein and chlorophyll of the swelled protoplasts appears to be a feature of senescence. Different culture conditions did not show any significant effect on the progress of senescence. There was a rapid decline in glutamine synthetase (GS, EC 6.3.1.2) and glutamate pyruvate transaminase (GPT, EC 2.6.1.2.) activities. In contrast, a glutamate dehydrogenase (GDH, EC 1.4.1.2.) activity increased progressively over the first two days. Especially, a marked temporary increase in the activity of NADH-GDH might compensate the decreasing GS activity.
