Abstract

Objective To observe the Caveoline-1 protein expression in spinal cord dorsal horn and dorsal root ganglia in diabetic peripheral neuropathy pain model rats at each observing point after STZ injection. Methods Fifty male SD rats were randomly devieded into two groups, 20 rats in control group were administrated with normal saline, and the other 30 rats were administrated with STZ(50 mg/kg) by introperitoneal injection. After one week of administration, the random blood glucose level from caudal vein was detected, and the rats whose random blood glucose were above 16.7 mmol/L were selected for further study, the index of paw withdrawal threshold(PWT) after two weeks of administration were tested, then the rat with PWT less than 4 g were selected. At each observing point, five rats were selected at the fourth, the sixth and eighth week after STZ injection to detect the Caveoline-1 protein expression in spinal cord dorsal horn and dorsal root ganglia with western-blot method. Results The Caveoline-1 protein expression in diabetic peripheral neuropathy pain model rats increased by (144.40±6.04)%, (105.50±5.12)%, (177.80±5.23)%, compared with the control group at four, six week and eight weeks after STZ injection in spinal cord dorsal horn, the differences were significant(P<0.05). The Caveoline-1 protein expression in diabetic peripheral neuropathy pain model rats increased by (128.60±6.51)%, (160.00±6.72)%, (120.10±6.18)%, compared with the control group at each observing point after STZ injection in dorsal root ganglia, the differences were significant(P<0.05). Conclusions The chronic pain caused by diabetes nerve damage can induce the increased amount of the expression of Caveolin-1 on spinal cord dorsal horn and dorsal root ganglia.
