Abstract
Three polypeptide cardiotoxins, the sputa cardiotoxin A (SCA), sputa cardiotoxin B (SCB), and sputa cardiotoxin C (SCC) have been isolated from the venom of Malayan cobra ( Naja naja sputatrix). The cardiotoxins were purified to electrophorectic homogeneity by successive column chromatography on SP-Sephadex C-25 ion exchanger and Sephadex G-50 gel media. The LD 50 (intravenous injection) values of the purified cardiotoxins were found to be 1.0 μg/g (SCA); 1.1 μg/g (SCB); and 1.05 μg/g (SCC). All three cardiotoxins exhibited direct lytic effect on human and guinea pig erythrocytes and caused complete arrest of the rat's atrial preparation. The molecular weight of the cardiotoxins was estimated to be 7000 by gel filtration chromatography and sodium dodeyl sulfate-polyacrylamide gel electrophoresis. Amino acid compositions of the three cardiotoxins are similar to those of other cobra venom cardiotoxins.
