Abstract

When small amounts (1.0 μg/g body weight) of diethylstilbestrol (DES) were given subcutaneously to pregnant mice daily from Day 13 to 18, low but significant yields of the lung and ovarian tumors were induced in the offspring. Lung tumor frequency (tumors/lung) was about one tenth that induced by the maximum tolerated dose (20 μg/g) of 7,12-dimethylbenz(a)-anthracene (DMBA). However, adenocarcinoma was not induced in the vagina, even when DES was given continuously as happened in humans and during the embryonic stage which is comparable to the stage of high sensitivity to DES in humans. DES showed marked teratogenicity. Almost all female offspring developed a urogenital anomaly (persistent urogenital sinus). In some cases, a firm, smoothly surfaced solid tissue bar was observed in the vagina. DES, as did DMBA, induced hypogenesis of male and female gonads.
