Abstract

This chapter deals with applications of mass spectrometry (MS) to analyze and structure elucidation of carbohydrates. The chapter includes routine methods such as gas chromatography–liquid chromatography–mass spectrometry (GC-LC-MS) analysis of volatile carbohydrate derivatives such as partially methylated alditol acetates for linkage analysis; high-performance liquid chromatography separation of oligosaccharide mixtures coupled with electrospray ionization tandem mass spectrometry; matrix-assisted laser desorption/ionization, time-of-flight mass spectrometry for profiling and structure analysis; and some advanced mass spectrometric methodologies created to determine the absolute configuration of monosaccharides or anomeric configuration of carbohydrate units in oligosaccharides available in trace amounts and to separate similar oligosaccharide molecules in a gas phase.
