Abstract

We studied the anatomy and histochemistry of Cocos nucifera leaflets aiming to improve the knowledge about the species and give future support to the research on coconut-foliar pathogens interaction. Fragments of the middle region of the leaflets were collected and used in this study. This material was processed for analysis by light microscopy and scanning electron microscopy. For histochemical study, transversal sections of fresh material were subjected to various histochemical tests. The anatomical analyzes revealed the presence of uniseriate epidermis, coated with plate-like epicuticular waxes. Tetracytic stomata and stellate trichomes were mainly observed on the abaxial face of epidermis. The dorsiventral mesophyll showed 2-3 layers of palisade, 4-5 layers of spongy parenchyma and collateral vascular bundles surrounded by sclerenchymatous pericycle. Idioblasts containing calcium oxalate crystals or phenolic compounds were also observed in the mesophyll. The anatomical and histochemical results observed in leaflets of C. nucifera may be involved in the interaction coconut-leaf pathogens.

O coqueiro (Cocos nucifera L.) pertence à tribo Cocoeae, subfamília Arecoideae e à família Arecaceae (Palmae) (Gunn, 2004). Como objetivo, descrever anatomicamente os folíolos de C. nucifera, utilizando microscopia de luz, métodos histoquímicos e microscopia eletrônica de varredura; visa, ainda, contribuir para o melhor conhecimento da espécie e da família Arecaceae e fornecer subsídios para futuros estudos de interação coqueiro/patógenos foliares. Para a análise estrutural dos folíolos em secção transversal, os fragmentos foram fixados em solução contendo 2,5% de glutaraldeído e 4,0% de formaldeído, diluídos em tampão Pipes a 1%; posteriormente, realizou-se a desidratação em série alcoólica além de inclusão em resina UNICRYL ; após a polimerização da resina o material foi seccionado transversalmente com navalhas de diamante (Diatone) a uma espessura de 0,5 μm, em ultramicrótomo (Reichet Ultracut S ). Os dados relativos à microscopia de luz foram obtidos e registrados digitalmente em microscópio Axioplan Zeiss, equipado com câmera ZVS – Zeiss, acoplado ao sistema AnaIySIS /Soft Imaging System de processamento digital de imagens
