Abstract

The vast majority of cellular proteins are degraded by the ubiquitin–proteasome pathway. One of the first-characterized and well-studied signals that targets proteins to this pathway involves the presence of ‘destabilizing’ amino acid residues at the amino terminus of proteins (the ‘N-end rule’). Because most of the proteins present within cells do not bear ‘destabilizing’ residues at their N-termini, the physiological significance of this pathway remains elusive.
