Abstract

From acid ethanol extracts of adult Drosophila melanogaster a highly cationic growth factor, CalGF, with mol. wt below 400 has been isolated. It contains no amino acids and supports the proliferation of the cell line KcHP at concentrations as low as 3ng/ml in a chemically defined medium ZO. Using CalGF supplemented ZO it is shown by single depletion experiments that 17 amino acids, 4 vitamins, nucleosides, oxocarboxylic acids and salts are strictly essential for the clonal proliferation of KcHP. Malic acid, carnitine, glutathione, lipids and 4 vitamins are each highly stimulatory. Using this information a likely minimal medium, ZR, is formulated.
