Abstract
The strain S3299-1 was identified from the Bacillus thuringiensis isolate (isolate No: 20080316S3299) that was isolated from soil samples collected from Diaoluoshan Tropical Rainforest Reserve in Hainan Province of China in 2008. Bt strain S3299-1 synthesizes the typical bipyramidal crystals during the sporulation that are lethal to Plutella xylostella larvae. PCR-RFLP analysis indicated that the cry-type of S3299-1 be high similar with that of Btk model strain HD 73. The inclusion proteins were highly expressed with the molecular weight of around 130 kDa illustrated by SDS-PAGE. Furthermore, larvicidal assay showed that protein inclusions of cry1Ac31 (Accession number GU446674) expressed in E. coli cells exhibited high toxicity against Plutella xylostell a larvae (LC 50 of 2.204 ng/mL), however, the mortality was less than that of toxin expressed in wild Bt strain (LC 50 of 1.504 ng/mL).
